The team found the potential to reverse the biological phenomenon of aging in skin cells through PDK1 inhibition model

Amorepacific R&D Center, together with the Korea Advanced Institute of Science & Technology (KAIST), developed an original technology that reverses the aging process in human dermal fibroblasts. The system’s biology research was conducted in collaboration with a research team led by Professor Cho Kwang-hyun of the Department of Bio and Brain Engineering at KAIST. The result of the research was published in the online edition of an internationally renowned scientific journal, PNAS, on November 23. (The research article is titled “Inhibition of 3-phosphoinositide–dependent protein kinase 1 (PDK1) can revert cellular senescence in human dermal fibroblasts”.)

In this industry-university research collaboration, Amorepacific R&D Center and Professor Cho’s team developed a signaling network model for aging skin cells. The joint team ran simulations to analyze the model and discovered a core f that reverses aging in old cells and converts them into younger cells. Using this factor, the team managed to develop an original technology for reverse aging.

These findings are meaningful in that we have found the potential to reverse the biological phenomenon of aging in skin cells that was previously considered irreversible. When we regulated the activity of PDK1 (3-phosphoinositide–dependent protein kinase 1), the core factor that reverse aging, in the artificial model of aged skin, we found that PDK1 inhibition not only reverses the reduction in collagen synthesis, but also helps recover the skin’s ability to regenerate. It is expected that we can leverage this reverse aging technology to not only predict and suppress the progress of age-related diseases, but also to prolong the healthy years of our lives.

Park Won-seok, Director of Research & Innovation Center at Amorepacific R&D Center said, “The joint research enabled us to build an aging signal network model and an artificial model for aged skin, with which we saw the potential to reverse the aging process in the skin.”

Based on the results of the studies, Amorepacific R&D Center is developing cosmetics that reduce wrinkles by extracting a core ingredient in Camellia japonica seed extract that controls aging in human skin.

Some adverse reactions to cosmetic procedures occur more often in patients with darker skin types. Patients should be counseled on appropriate cosmetic procedures for their skin type, according to Cheryl M. Burgess, M.D., at the Skin of Color Update virtual meeting.

Whether or not dermatologists perform cosmetic procedures, it is important that they understand the spectrum of adverse reactions that can occur in patients with skin of color because they may see patients seeking care because of a com- plication, said Cheryl M. Burgess, M.D., at the Skin of Color Update virtual meeting. “With the advent of medical spas and the cross-over of various specialists into cosmetic procedures, I have seen an upsurge in the number of patients with Fitzpatrick skin types IV, V, and VI who come to me because they experienced an adverse event following a treatment involving a laser or light-based device, filler or neuromodulator injection, or chemical peel,” says Dr. Burgess, founder and president, Center for Dermatology and Dermatologic Surgery, Washington, D.C.

She described the types of reactions that are seen most often in skin of color and offered recommendations for minimizing risks and achieving patient satisfaction. Summarizing her top tips, Dr. Burgess says, “Careful patient selection is important for achieving the desired out- come with any cosmetic procedure, and thorough counseling should be provided at consultation on procedures that are appropriate for the individual’s skin type. Performing a test spot may be useful to determine potential adverse events with lasers and chemical peels, and laser parameters may need to be tweaked for increased safety.”

She emphasizes that it is best to start conservatively and recognize that achieving the desired outcome may require multiple sessions.

Laser and light systems

First and second-degree burns are among the most common complications seen in skin of color patients who undergo cosmetic procedures involving a laser, and they are particularly a problem with the use of intense pulsed light devices. The burns are easily recognized by their rectangular shape, Dr. Burgess says.

Postinflammatory hypopigmentation can occur after hair removal using the short pulse 1064-nm Q-switched Nd:YAG laser whereas hyperpigmentation was more common in the past when the procedure was performed with the 532-nm systems.

“The longer 1064-nm wavelength avoids hypopigmentary changes since it bypasses the melanin in the epidermis. Its absorption deeper in the hair leads to thermal destruction of the follicle, which makes the longer wave Nd:YAG laser ideal for hair removal in skin of color,” Dr. Burgess says.

Whenever performing a laser procedure in a new patient with skin of color, however, Dr. Burgess advises performing a test spot and using a lower fluence.

“Manufacturer-recommended parameters are based on clinical trials that enrolled mostly patients with Fitzpatrick skin types I-III and some patients with skin type IV, but rarely any- one with darker skin,” she says. “When patients understand the test is being done for their safety, they are willing to accept the delay.”

Filler reactions

Safety data from premarketing clinical trials with various fillers did not show any increased risk of pigmentary changes or hypertrophic or keloidal scarring in patients with skin of color, although the eligibility criteria for these studies excluded patients with a history of keloids. As in patients with lighter skin types, bruising is the most common adverse event associated with filler injections in skin of color. Hyperpigmentation can also occur, particularly with the use of a serial puncture technique and faster injections times.

“Filler injections in skin of color patients are best done slowly using a linear threading technique,” Dr. Burgess says.

Neuromodulator injections

Dr. Burgess notes that Asian patients, particularly, request treatment for crow’s feet and glabellar frown lines. Injections into the forehead and periocular region in these patients requires care to avoid the development of eyebrow or eyelid ptosis that will decrease the palpebral aperture.

“As people age and the laxity of the forehead and upper eyelid skin becomes more

apparent, a reduced dose or avoidance of botulinumtoxinA injections is recommended to avoid eyebrow ptosis,” Dr. Burgess says. Chemical peels

Superficial chemical peels using salicylic acid, lactic acid, malic acid, or glycolic acid can be performed safely in skin of color. The use of medium and deep peeling agents, however, risks dyschromia from rapid skin exfoliation that leads to a deeper exfoliation to the lower levels of the epidermis and melanocytes with subsequent inflammation and ultimately hyperpigmentation.

Dr. Burgess says that neutralization of the acid is a technique used in medical spas to minimize risk in skin of color. However, a neutral pH is no longer an acid strength to perform chemoexfoliation.

“Chemical peels at lower pH require observation of the skin’s reaction and erythema. The end point for a peel isn’t always ‘until it frosts’ in skin of color,” Dr. Burgess says

Source – https://timesofindia.indiatimes.com/

Real beauty is reflected in your skin. If you want a clearer, younger looking skin, non-surgical cosmetic treatments and some maintenance after that can let you achieve immediately visible and long-lasting results. You don’t have to always undergo cosmetic surgery to improve appearance and reduce the signs of ageing. In fact, you can choose non-surgical cosmetic treatments to reduce wrinkles, plump up your lips, smoothen the surface of your skin. We spoke with Dr. Anup Dhir Senior Consultant, Cosmetic Surgeon, Apollo Hospital, Delhi to know more about non-cosmetic procedures that can bring a drastic change in your appearance and here’s what the doctor had to say.
Non-surgical cosmetic treatments: Broadly, these treatments are of three types. We can use laser and other energy based devices like radiofrequency and ultrasound, we can do injectables like botox, fillers microfat and PRP, or we can do time tested older procedures like chemical peels, dermabrasion and microneedling to rejuvenate the skin.

Laser skin resurfacing for wrinkles is very commonly done with carbon dioxide or erbium fractional laser and helps by removing the top layer of the skin and making it look younger. Laser hair reduction is done with lasers for facial and body hair and normally six sittings are needed at monthly intervals.
Radiofrequency energy devices like thermage, exilis, e matrix etc. and ultrasound devices like HIFU help in formation of new collagen under the skin by directing energy at a particular level under the skin.

Botox injection for wrinkles -This injection is very commonly used for treatment of facial wrinkles and weakens the muscles which cause wrinkles. It has to be repeated after 4-6 months.

Anti-wrinkle treatment by fillers is again a very popular procedure in which hyaluronic acid fillers are injected into scars and wrinkles. The resulting improvement lasts for 9-15 months.

Fat injection for scars and wrinkles are the gold standard for rejuvenation and in this, your own body fat is sucked, processed and injected in the facial wrinkles. Microfat and nanofat are the types of fat which can be used. The fat has its own stem cells and they help to rejuvenate the skin and improve texture and; help in face lift and rejuvenation of hands also.

PRP skin rejuvenation is done by taking your blood and making platelet rich plasma from it and this is injected in the facial skin and it can also be used in the scalp to reduce the hair fall and help in regrowth.

Dermabrasion involves taking the outer layer of skin with a diamond roller under a local anesthetic and the new skin on healing has less scars. The purpose of surgical dermabrasion is to help diminish the appearance of deeper scars and skin imperfections and smoothen the skin.

Chemical peels use a chemical solution to smoothen the texture of your skin by removing the damaged outer layers. It is one of the least invasive ways to improve the appearance of your skin. Superficial peels with fruit acids like glycolic acid etc. are also called lunchtime peels as they have hardly any down time and can give quick results. As sun exposure, acne, or just getting older can leave your skin tone uneven, wrinkled, spotted or scarred, these peels can help these conditions. They also helps to whiten the skin.

Dermaroller and mesotherapy can be done together and while doing it, a skin roller is used on the skin and mesotherapy solution is injected to improve acne scars and treat the wrinkles. Dermaroller can also be used with PRP and nanofat to rejuvenate the skin.

Finally, anti ageing diet rich in antioxidants for healthy skin has a big role to rejuvenate from within. Fruits, multi coloured vegetables, fish, plenty of water etc. are known to make your skin look younger.

Proper care and maintenance of skin is needed to achieve long lasting results. Don’t forget daily use of sunblock SPF30 or above to save your skin from premature ageing due to UV radiation in sun rays. However, you need to consult a cosmetic surgeon to find which method is best suited for your skin type.

Aging decreases our body’s capacity for synthesizing new proteins, and as a result collagen levels can fall by as much as 75% by the time a human being reaches 80 years of age.

Experts in several biological fields such as longevity or dermatology have suggested that supplementing with hydrolyzed collagen (HC) can delay some of the hallmark traits we associate with aging such as hair loss, wrinkling of the skin, joint and muscle pain, and skeletal brittleness.

Here’s a look at the literature on how hydrolyzed collagen supplements can be good for not just bones, joints, and skin, but also for attenuating the effects of certain diseases like osteoarthritis, diabetes, and inflammation.

Hydrolyzed collagen for bones

As one of the sturdiest protein structures owing to its triple-helical shape, HC can benefit bone health in a number of ways, both as an attenuator of bone diseases like osteoarthritis, and as a fortification for bone cells.

“The benefits of hydrolyzed collagen for osteoporosis prevention go beyond the effect of a simple protein supplementation,” write the authors of an in vitro study which found that bovine-derived HC stimulated the growth of bone cells like osteoclasts and osteoblasts, while increasing mineralized bone-matrix formation–findings which corresponded with an in vivo analysis of post-menopausal woman with osteoarthritis.

Rhonda Patrick Ph.D. highlights on her blog that the importance for collagen in bone health can be summarized by the fact that certain genetic mutations in collagen-producing genes can lead to misshapen and more easily fractured bone structures.

Bone mineral densities were increased in post-menopausal woman with reduced bone density, notably around the neck and spine, after a period of supplementing with a hefty 5 grams of HC accompanied by 500 to 800 milligrams of calcium and 400 – 800 IUs of vitamin D.

Contradictory research done on a similar group of women with osteopenia, a condition characterized by bone-density loss, found no gains in bone metabolism. However some of the woman were deficient in calcium, and others were overweight, both conditions that are proven to negatively affect bone health.

CHECK OUT: Bone Broth Strengthens Immunity, Body, and Joints, And You Don’t Need To Spend A Bunch To Make It

While this muddies the waters in the discussion on bone health when supplementing with HC, it may have revealed that simply accompanying it with a calcium supplement is enough to see the similar benefits experienced by the individuals in the other studies.

Hydrolyzed collagen for skin health

The ultimate reason to describe HC as an anti-aging supplement would be its effects on skin.

Type I and III collagen are most common in skin and have been shown in multiple studies to increase hydration levels, and reduce the appearance of wrinkles, crow’s feet, and numerous other decreases in facial elasticity.

“Knowledge of the role of collagen in the aging process over 30 years ago helped to establish the use of bovine collagen as a filling agent to temporarily replace collagen lost with age in soft tissue” write the authors of a broad review on dermal aging, noting another study that found collagen production decreases on average by about 1% per year.

Skin dryness, crow’s feet, wrinkles, and overall skin elasticity improved by 7% after 4-8 weeks of supplementing with between 2.5 and 5 grams of HC, with a notably more significant result coming from the oldest women in the randomized controlled trial (RCT).

READ: Dreading a Dark Winter Lockdown? Think Like a Norwegian

It should be noted that collagen is the highest correlating factor with skin thickness, or a lack thereof, and if anti-aging is to be considered function over form, then skin less easily penetrated by UV rays, bacteria, and dust is perhaps more desirable for some individuals than younger looking skin.

Another RCT of women from 40-60 years of age who took 1 gram of HC along with vitamin C for 12 weeks showed that the HC-supplemented group had a 7.23-fold greater improvement in skin hydration at six weeks.

Studies suggest that chronic dehydration can accelerate the aging process, including signs of aging on the skin. If you struggle with dry skin, wrinkles, loss of facial volume and other symptoms associated with skin aging, rehydrating your skin is the first step to getting a healthy, youthful-looking complexion.

THE LINK BETWEEN DRY SKIN AND AGING

We know that dry skin accentuates signs of skin aging like lines and wrinkles, as less water retention contributes to less facial volume. Thus, lines, wrinkles and skin laxity are more noticeable on dry, dehydrated skin.

Additionally, dry skin is often also an indication of dehydration throughout the body, which can contribute to accelerated aging of all of the body’s organs, including its largest — the skin.

While there is a genetic component to dry skin, there are ways you can improve your skin’s hydration and overall health, despite your genetic skin type:

Drink plenty of water. Hydrating the skin starts from the inside out. It’s also important to make sure your body is getting enough water to perform daily functions properly and keep your skin looking healthy and glowing.

Avoid excessive alcohol consumption. Alcohol dehydrates your body, including your skin. This is why most people wake up the day after drinking with a dull, lackluster complexion. If you are going to indulge in a drink or two, be sure to drink water in between.

Use a barrier repair moisturizer. Barrier repair moisturizers like Zerafite Barrier Repair Moisturizer and Medature PSL Repair Moisturizer and EpiCeram (requires a prescription) help the skin to replenish and repair its natural skin barrier so that it is able to retain water, rather than allowing it to evaporate off the skin.

A good barrier repair moisturizer should contain a 1:1:1 ratio of ceramides, fatty acids, and cholesterol to mimic the skin’s natural barrier components. Look on the label for “MLE technology” or “PSL technology” to know that the moisturizer has the proper lipid ratio.

BOTTOM LINE ON HYDRATING YOUR SKIN

Dry skin is a sign of dehydration throughout the body and is one of the four barriers to skin health. While some people are genetically predisposed to having dry skin, the right skincare routine — including a good barrier repair moisturizer — and lifestyle habits like drinking enough water and avoiding excess alcohol and caffeine can help to reverse dry skin and help you maintain a healthy, youthful-looking complexion.

A new tool has been developed that can effectively predict a person’s chronological age based on a microbiome sample. The tool, developed in collaboration between UC San Diego and IBM researchers, is most accurate at predicting a person’s age when using a skin microbiome sample.

A number of intriguing studies are beginning to investigate links between aging and the billions of microbes that live within and on us. Researchers from Singapore last year experimented with gut microbiome transplants between old and young mice, with the results suggesting an aging microbiome may alter its bacterial populations to balance out the more systemic deficits of aging in an organism.

This new research set out to use modern machine learning techniques to investigate whether the human microbiome could be used as an indicator of age. Three different human microbiome populations were explored in the study – skin, mouth and gut. Mining data from 10 previously published studies, the new research gathered almost 9,000 microbiome samples from subjects aged between 18 and 90 years.

Interestingly, the model found the skin microbiome to be the most accurate predictor of chronological age, correctly estimating age within a window of 3.8 years. The oral microbiome was similarly accurate, with correct estimates within 4.5 years. The gut microbiome was the least accurate, found on average to only be correct within a window of 11.5 years.

The skin microbiome is sampled using swabs from either the hand or forehead, and the research suggests both locations offer equally accurate results. The skin microbiome samples were also the most consistent across different cohorts and geographical locations, suggesting our skin microbes are quite similar no matter where we live or what we eat.

The researchers hypothesize the reason the skin microbiome is the most accurate predictor of chronological age is most likely due to general age-related physiological changes in skin conditions. As most people age their skin becomes drier and decreases serum production, and these changes can be explicitly reflected in alterations to superficial microbiome populations.

“The human microbiome is constantly evolving and has been observed to change with age,” three of the researchers write in an editorial accompanying the newly published study. “The presence of unusually early microbiome aging patterns, relative to chronological age, could potentially signal altered susceptibility for age-related diseases. Conversely, a ‘young’ microbiome might offer clues on how to decelerate the aging process.”

The current research is based on microbiome data from subjects in the United States, China, the United Kingdom, and Tanzania. More data, and larger studies, will be necessary to better home in on the microbiome differences between various geographical and ethnic populations. However, the researchers are confident the particular universality of the skin microbiome may help home in on a general relationship between microbes and aging that unifies humans across the globe.

“This new ability to correlate microbes with age will help us advance future studies of the roles microbes play in the aging process and age-related diseases, and allow us to better test potential therapeutic interventions that target microbiomes,” says Zhenjiang Zech Xu, co-senior author on the study.

Source: news-medical.net

Tummy tuck surgery (abdominoplasty) yields high patient satisfaction and improved quality of life in patients who are overweight or obese – despite a substantial risk of complications, reports a study in the October issue of Plastic and Reconstructive Surgery®, the official medical journal of the American Society of Plastic Surgeons (ASPS).

An “overwhelming majority” of overweight/obese patients are happy with the results of abdominoplasty, according to the study by Dennis C. Hammond, MD, and colleagues of Partners in Plastic Surgery of West Michigan, Grand Rapids. They write, “A real quality of life improvement can be obtained by offering body contouring even in the face of obesity, with the caveat that the risk of minor postoperative complications is high.”

Abdominoplasty is an effective procedure to improve the appearance of the abdomen. However, this and other body contouring procedures have historically been discouraged in overweight or obese patients. That reflects concerns that increased body mass index (BMI) may increase the risk of wound healing problems and other complications.

The researchers analyzed the outcomes of tummy tuck surgery in 46 overweight/obese patients over a 12-year period. The patients were 41 women and five men, average age 49 years. All had a BMI of 25 or higher, with an average BMI of 32. (A BMI of 25 or higher is considered overweight, while a BMI of 30 is the cutoff point for obesity.)

Eighty percent of patients underwent abdominoplasty, most often including a procedure to restore weakened or separated abdominal muscles. The remaining 20 percent had a procedure called panniculectomy to eliminate excess, “hanging” abdominal fat and skin. The researchers analyzed the outcomes of surgery, including complication rates and patient-rated outcomes.

Nearly half of patients had some type of complication. About 39 percent had minor complications, requiring office procedures or antibiotics. About nine percent of patients had major complications requiring a return to the operating room – mainly due to wound healing problems and/or fluid collections.

Thirty-six patients completed follow-up surveys an average of 15 months after their procedure. Ninety-four percent of patients were satisfied with the results of abdominoplasty/panniculectomy, while 97 percent stated they would choose to have the procedure again. Ninety-seven percent of patients said the procedure had improved their quality of life. Nearly half of patients said they lost additional weight after surgery.

“Abdominoplasty and panniculectomy in the overweight and obese patient presents as a surgical decision-making challenge for the treating surgeon,” Dr. Hammond and coauthors write. Due to their increased risk of complications, patients are commonly advised to lose weight before undergoing body contouring surgery.

The researchers note, “Even with weight loss, the excess skin and fat…will not completely recede and can still present as an impediment to normal function and exercise.” While acknowledging the increased risks, they offered abdominoplasty or panniculectomy to overweight/obese patients “in an attempt to relieve the discomfort and physical effects of the excess skin and fat and offer the potential to jumpstart a weight loss process.”

The authors believe their results support this strategy. Although complications were frequent, most were minor and readily manageable, and many patients lost more weight after surgery. Dr. Hammond and colleagues conclude: “[E]ven in the face of this elevated complication rate, patient satisfaction is overwhelmingly high, making body contouring procedures in this patient population an acceptable option in appropriately selected patients.”

SEOUL — Cosmax, a cosmetics original development manufacturing company in South Korea, has developed an anti-aging cosmetics material called “Strain CX” using microbiome, the genetic material of all the microbes that live on and inside the human body. Strain CX is a composite of microbes with anti-aging characteristics found in the skin of young women.

In the cosmetics industry, microbiome specifically refers to a group of microbes which are known to improve the conditions of internal organs and skin. Microbiome has received attention as a material for next-generation skincare cosmetics, but most cosmetics products that utilize microbiome research technology have used intestinal lactobacillus or milk lactobacillus.

Cosmax promised to release a new lineup of cosmetics that would enhance the anti-aging capability of products targeting fall and winter seasonal markets.

“While all other companies were focusing on the balance of microbiome and the relationship between aging and microbes, we thought that we should approach the skin,” Cosmax organic materials lab head Lee dong-geol said. “Our research performance is based on finding out exactly what our customers want after meeting with them several times.”

“Because skin microbiological research is much higher in the level of research analysis than intestinal microbial research, I have spent the last five years just making a platform for analysis,” Lee said, adding his team found the correlation between anti-aging microbes and skin aging and divided them.

Strain CX can greatly enhance moisture activity and collagen activation, he said. Cosmax promised to release a new lineup of cosmetics that would enhance the anti-aging capability of products targeting fall and winter seasonal markets.

Strain CX was first adopted by Dr. Jart+, a cosmetics brand, to release a moisturizing essence. The product has become the top-selling merchandise at Olive Young, a health and beauty drug store, this summer. Dr. Jart+ will release a new lineup of cosmetics featuring a more concentrated dose of Strain CX.

“While the moisture biome concentrates on the moisturization capability of cosmetics, a new lineup featuring the concentrated dose of the anti-aging element, which is the core of the Strain CX, will be released as autumn and winter come,” Lee said.

It is a great idea to include banana in your beauty regime and enjoy the benefits of the fruit which is rich in potassium and vitamin A, B, and C!

Banana is one of the most consumed fruit as it is easily available across the globe. But this fruit is not only tasty to eat, it also has multiple benefits for one’s health, skin, and even hair. While eating banana helps boost energy, its puree is an excellent way to get amazing skin and silky and smooth hair. It is also useful to fight off premature aging of the skin. Hence, it is a great idea to include banana in your beauty regimen and enjoy the benefits of the fruit which is rich in potassium and vitamin A, B, and C.

Skincare benefits:

Skin moisturizer

“Bananas are the secret to a well-nourished skin as it helps keep it hydrated. It also helps moisturize dry skin, making it soft and supple. Vitamin A in banana restores the skin’s moisture and smoothens out the rough texture by healing dry and parched skin,” says Plabita Sharma, skincare expert, The Body Shop.

Bananas contain high level of antioxidants and act as nature’s Botox, preventing fine lines and wrinkles from forming.

Treats acne

Bananas are loaded with many nutrients like vitamin A, zinc, and manganese, which are anti-inflammatory. Rubbing a banana peel on your face acts as a blemish banisher, and also helps in treating acne.

Dandruff can be caused by scalp irritation, dryness, as well as fungal and bacterial pathogens. “Applying banana mask to your scalp can add moisture (cutting down on dryness) and also rid your scalp of microscopic offenders causing dandruff,” says Sharma adding that for centuries, banana peel, leaf, flower and fruit have been in used to treat various ailments as the extracts of different parts of the banana have antioxidant and antibacterial properties.

Banana may help smoothen frizzy hair as it has high silica content. Silica is absorbed by our body to produce collagen, a protein that is a building block of bouncy and beautiful hair.

Bananas are a great way to get soft, smooth, manageable as well as moisturised hair. Because of its rich potassium, vitamin, calcium, and carbohydrates content along with natural oils, bananas provide nourishment to the hair and makes it smooth and shiny.

Antioxidants in banana can also infuse your scalp and hair with a stronger defense system, and decrease oxidative stress — a major reason for hair to look brittle and aged. Using banana overtime can make hair follicles stronger, and, as a result, grow longer.

Liposuction is that the gold commonplace methodology to produce permanent spot reduction and contour improvement in areas laid low with excess fatty deposits. Your fat cells are distributed throughout your body early in life, as determined by your biology. Fat cells cannot migrate or multiply, and once they’re removed, they cannot return. Therefore, any contour improvement achieved with liposuction is a permanent improvement and can persist, not with standing a patient gains weight.
Suction-assisted reconstructive surgery typically takes an hour or 2; however the time needed could vary significantly — from thirty minutes to many hours, betting on the area and quantity being suctioned.

Liposuction is a cosmetic procedure that’s accustomed trim and reshapes hassle areas of the body that diet and exercise can’t appear to focus on. Liposuction removes excess fat deposits, leading to an additional ideal body contour.

To begin the procedure, this clinic makes a tiny low incision, simply massive enough to permit the insertion of a hollow tube known as tubing. The alternative finish of the tubing is hooked up to a machine that makes a powerful vacuum.
Source: eurekalert.org

The cell polarity protein Par3 controls mechanic changes in the skin and plays an important role in cell division. Malfunction can lead to DNA damages. The balance of the system is of great importance: while too much differentiation leads to loss of stem cells and therefore premature aging, too many cell divisions can be a cause of skin cancer. The new study by a team around Sandra Iden about how polarity regulators control cellular mechanics in the skin was now published in Nature Communications.

The skin serves as a crucial barrier to the outside world. Its task is not only to keep pathogens or toxic chemicals out of the body, but also to keep water inside and maintain hydration. In order to function properly, the skin epidermis constantly needs to keep a balance of cells it sheds off and new cells that replenish the lost ones. The skin epidermis is made up of several layers with different functions. Skin stem cells are responsible for the self-renewing capacity of the skin.

In a previous study, the researchers showed that inactivation of the polarity protein Par3 resulted in a decline of stem cells, impaired skin homeostasis and premature skin aging. Back then, however, the underlying mechanisms remained unclear. ‘We were now able to show that Par3 has a direct influence on the homeostasis of the skin by controlling the mechanical properties of keratinocytes, the main skin epithelial cell,’ said leading scientist of the study Dr Sandra Iden. The polarity protein Par3 controls the mechanical properties of the main skin epithelial cells, called keratinocytes. It has functions that are conserved from worms and flies to mammals. Par3 also regulates barrier function and cell division orientation.

The recent work started with two separate approaches. One of the first authors, Martim Dias Gomes, said: ‘We realized that inactivating Par3 leads to failures in cell divisions, resulting in DNA damage responses.’ At the same time, his colleague and co-first author Soriba Letzian was working on another project: ‘We challenged mouse skin with UV light – but observed an unexpected DNA damage response already in absence of the harmful light, when Par3 was missing. That was the moment we realized that the DNA damage response and the aberrant cell divisions might be tightly linked,’ he said. Based on these results, they teamed up and together examined possible causes of these mitotic failures.

As they now show, Par3 is an important regulator of contractility of keratinocytes, which is required to maintain the accuracy of cell division events. The absence of Par3 led to mitotic errors, causing an alert signal and a cascade of DNA damage responses that then fuelled premature differentiation, and potentially the skin stem cell decline. These findings were surprising, as during the development of epithelial tissues Par3 was considered to serve tissue function rather through orienting mitotic spindles.

These new findings thus revealed that core polarity proteins like Par3 steer mechanochemical networks essential to keep a healthy self-renewal capacity. ‘We are glad that we were able to contribute a piece of the puzzle of how the skin epithelium is maintained intact, and hope that this will serve future medical applications,’ Sandra Iden concludes.

Cosmetic companies promise to make you look younger, so long as you buy a multitude of their products and continually use them for the rest of your life. Humankind’s quest for the fountain of youth has often come at great expense both financially and morally, yet there has been some modicum of scientific information gleaned from our vanity driven endeavors over the years. Now, investigators at Johns Hopkins University may have just uncovered the mechanisms driving the rejuvenation process that dermatologists have exploited for many years.

Interestingly, the researchers discovered that laser treatments and the drug retinoic acid share a common molecular pathway. Moreover, that pathway—which lets skin cells sense loose RNA molecules—is also turned up in mice when they regenerate hair follicles. Findings from the new study were published recently in Nature Communications through an article titled “Noncoding dsRNA induces retinoic acid synthesis to stimulate hair follicle regeneration via TLR3.”

“Understanding the biology behind how cellular damage can lead to this type of regeneration can harness a new generation of therapeutics,” explained senior study investigator Luis Garza, MD, PhD, associate professor of dermatology at the Johns Hopkins University School of Medicine.

Researchers have known for decades that mice—unlike humans—can regenerate hair follicles after a deep wound. Recent studies by Garza and others found that loose pieces of RNA, called self-noncoding double-stranded RNA (dsRNA) can spur this regeneration. They hypothesize that this may be because dsRNA is released by damaged cells at the site of a wound. Garza and his colleagues were curious whether dsRNA also played a role in skin rejuvenation treatments such as laser therapy, microneedling, and facial abrasion, which all involve temporary damage to skin cells. Although these treatments are well-established among dermatologists, researchers haven’t been clear why they work.

Garza and his colleagues analyzed the expression levels of genes in each sample and discovered that genes involved in sensing dsRNA as well as genes involved in producing the skin’s natural retinoic acid were all expressed at higher levels after the laser treatment. Next, the researchers treated isolated human skin cells with loose dsRNA—mimicking the effect of laser treatment. The amount of retinoic acid inside the cells increased by more than tenfold. Commercially produced retinoic acid is already used to treat acne, wrinkles, and sunspots.

“It’s not an accident that laser rejuvenation and retinoic acid have both been successful treatments for premature aging of the skin from sun damage and other forms of exposure,” Garza noted. “They’re actually working in the same molecular pathways and nobody knew that until now.”

To further strengthen and understand the connection, the researchers turned back to mice. They knew that in both mice and humans, a protein called toll-like receptor 3 (TLR3) senses dsRNA. When Garza’s group engineered mice to lack TLR3, the animals could no longer regenerate hair follicles after a wound. But when the researchers gave these mice retinoic acid, they regained the ability to regenerate the follicles. The results point toward a pathway involving TLR3 that senses double-stranded RNA and turns up the synthesis of retinoic acid.

“We show that self-noncoding dsRNA activates the anti-viral receptor TLR3 to induce intrinsic retinoic acid (RA) synthesis in a pattern that predicts new hair follicle formation after wounding in mice,” the authors wrote. “Additionally, in humans, rejuvenation lasers induce gene expression signatures for dsRNA and RA, with measurable increases in intrinsic RA synthesis.”

“In retrospect, it makes a lot of sense because retinoic acid is already a mainstay of wrinkle reduction and nobody knew what turned it on,” Garza remarked. “Now we know that damage leads to dsRNA, which leads to TLR3 activation and retinoic acid synthesis.”

The findings could lead to novel strategies to reduce wrinkles and sunspots by combining retinoic acid and laser treatments in new ways, Garza says. And they could also lead to ways to regenerate hair follicles, as mice do when there’s an increase in dsRNA after a wound.

“After a burn, humans don’t regenerate structures like hair follicles and sweat glands that used to be there,” said Garza. “It’s possible in light of these new findings that double-stranded RNA may be able to improve the appearance of burn scars.”

“These results demonstrate a potent stimulus for RA synthesis by non-coding dsRNA, relevant to their broad functions in development and immunity,” the authors concluded.

Nebraska ranks 8th in the nation for new cases of melanoma, a serious type of skin cancer. Spending time in the sun increases your risk of skin cancer and early skin aging. People of all skin colors are at risk for this damage.

We get sun damage to our skin by invisible ultraviolet (UV) radiation. Sunburn is a type of skin damage caused by the sun. Tanning also is a sign of the skin reacting to potentially damaging UV radiation.

You can lessen your risk by:

• Limiting your time in the sun, especially between 10 a.m. and 2 p.m. This is when the sun’s rays are most intense.

• Wearing clothing to cover skin exposed to the sun—such as long-sleeve shirts, pants, sunglasses, and broad-brim hats. Sun-protective clothing is available. (The FDA regulates these products only if they are intended to be used for medical purposes.)

• Using broad spectrum sunscreens with a Sun Protection Factor (SPF) value of 15 or higher regularly and as directed. (Broad spectrum sunscreens offer protection against both UVA and UVB rays, two types of the sun’s ultraviolet radiation.)

• Always reading the label to ensure you use your sunscreen correctly. Ask your doctor or nurse before using sunscreen on infants younger than 6 months. The best option is to keep infants younger than 6 months out of direct sunlight. When outside during the day, keep infants in the shade.

In general, the FDA recommends that you use broad spectrum sunscreen with an SPF of 15 or higher, even on cloudy days.

• Apply sunscreen liberally to all uncovered skin. Use it especially your nose, ears, neck, hands, feet, and lips (but avoid putting it inside your mouth and eyes).

• Reapply at least every two hours. Apply more often if you’re swimming or sweating. (Read the label for your specific sunscreen. An average-size adult or child needs at least one ounce of sunscreen to evenly cover the body. This is about the amount it takes to fill a shot glass.)

• If you don’t have much hair, apply sunscreen to the top of your head, or wear a hat. Use a hat that also covers your ears and back of your neck

• No sunscreen completely blocks UV radiation. So other protections are needed, such as protective clothing, sunglasses, and staying in the shade.

A dermatologist has revealed her top tips to protecting your skin from damaging city pollution, which can reduce antioxidants in skin and disrupt collagen production, aging skin as a result.

Shocking statistics released by the Department for Environment Food & Rural Affairs recently revealed that city pollution increases by a staggering 20 per cent on weekdays compared to weekends.

And now a skin expert has told FEMAIL that as well serious environmental implications and health consequences, pollution can have an ageing impact on our skin.

According to leading skincare clinic group sknclinics.co.uk only 23 per cent of Brits know how to protect their skin from harm, while 75 per cent of Londoners feel their commute has a negative impact on the condition of their skin.

Consultant Jinah Yoo revealed that deep cleansing at night to rid the pores of deep-set grime, as well as using SPF on a daily basis – even in the winter – and drinking eight glasses of water by 8pm, are all ways to keep your skin looking youthful.

Explaining how pollution can affect the skin, Jinah Yoo said: ‘There are several mechanisms by which air pollution can affect the skin.

‘Ozone, which is one of the major air pollutants, generates free radicals which leads to a reduction in the antioxidants in our skin, as well as disrupting collagen production and destroying the skin’s barrier. As a result, it promotes skin ageing.

‘With long-term exposure to air pollutants, you may see the signs of skin ageing which will appear as wrinkles, brown spots, uneven skin tone, dehydration and loosing elasticity of the skin.

‘Air pollutants also cause skin inflammation and increased pigment production.

‘What’s more, some of the air pollutants are small enough to penetrate into pores which can be trapped and cause acne-like spots.’

Advising how the ideal daily skincare routine should look, she continued: ‘There are four important daily skincare steps you should consider: deep cleansing, antioxidant application, moisturiser and sunscreen.

‘Firstly, deep cleansing at night will help to remove pollutants accumulated on the surface of the skin during the daytime.

‘After cleansing, application of an antioxidant cream containing vitamin C and E, which inhibit free radical formation, will help to delay the skin ageing process caused by extrinsic factors such as air pollution or UV.

‘However, choosing the right antioxidant and using it correctly is important. Vitamin C cream comes in various concentrations (5-20 per cent), it is best to start with a low concentration, then gradually increase the concentration as starting with high strength can occasionally cause skin irritation’.

Medical Director and specialist in Laser and Cosmetic Dermatology, at sk:n, Dr Hussein, shared his top daily skincare tips for keeping your face clean and clear in the city.

Keep an eye on the pollution forecast and air quality which will have a colour-coded map to show the areas of the UK worst effected by pollution.

Avoid spending unnecessary time near busy roads and traffic ingested areas when the map is coded in red, which is a ‘high’ air pollution banding, or purple, which signifies very high.

If you do find yourself unavoidable in a pollution hotspot, try to avoid touching your face during the day, and wash your face as soon as you get home.

Thoroughly cleansing your skin at night will help to remove pollutants accumulated on the surface of it during the daytime.

Exfoliating is a great way to remove the top layer of mostly dead, epidermal cells on the outer surface of the skin to expose fresher brighter skin.

Try an exfoliating cleanser that contains glycolic acid and glycerine to remove dead skin cells, clear pores, moisturise and rejuvenate the skin.

After a busy day avoid the temptation to use make-up wipes to cleanse your skin – they’re not a viable alternative to a proper cleanse and you could be doing more harm than good.

Make-up wipes can simply spread make-up, dirt and skin over your face, leaving irritated skin in their wake, and often contain ingredients such as alcohol or certain preservatives that will irritate more delicate skin types.

The scrubbing action that is often accompanied with their use can cause inflammation that, over time, can promote skin pigmentation.

After cleansing, application of an antioxidant cream containing vitamin C and E, which inhibit free radical formation, will help to delay the skin ageing process caused by extrinsic factors such as air pollution or UV.

Vitamin C cream comes in various concentrations (5-20 per cent), and it is best to start with a low concentration, then gradually increase the concentration as starting with high strength can occasionally cause skin irritation.

Keeping your skin well-moisturised will help to maintain an intact skin barrier, vital for protection against pollution.

Apply moisturiser morning and night after cleansing your face. You may wish to use a heavier moisturiser in the evening and a lighter one in the day.

Air pollution can affect everyone, however patients with underlying skin conditions with a broken skin barrier, such as eczema, may experience worsening of their eczema with exposure to air pollution.

Sun cream should be a key element of everyone’s daily skin routine, and not just in the summer.

Using sun cream which contains UV and visible light block will help to reduce signs of skin ageing.

It’s tempting to use more cosmetics to cover up any skin imperfections, however, by doing this it clogs your pores even more and triggers further outbreaks or can be what caused one in the first place.

Going cold turkey isn’t always a realistic option more some people so instead why try going one day a week with no make-up or using mineral cosmetics instead.

The worst culprits for causing breakouts are heavy use of foundation – look for products that are non-comedogenic, sheer, and fragrance-free.

When skin is dehydrated, it will be dull, lack moisture and therefore lustre.

The phrase ‘8 by 8’ is useful at keeping track of how many glasses of water you consume before the end of the day; simply drink eight glasses of water by 8pm, a minimum of 2 litres, and you’ll have had your fill for the day.

Whilst maintaining a good daily skincare routine will help protect your skin from damage, there are a variety of treatments that can help improve skin pigmentation and tone whilst rejuvenating the skin – including hydrofacials and laser treatments.
